For easy OD calibration I have labeled 20ml vials of standard OD made from impranil in water.
I used blank water during the OD calibration thinking to blank the media when setting up the experiment. The ability to blank the pure media from the GUI is gone now.
I think this setup is not ideal because it requires recalibrating OD with changes in media type, rather than just setting the blank. Perhaps I should not have used the water 0.0 in the orignal setup? When i tried to use the 0.0 standard as just a regular OD point and not the blank, i got an error for division by 0.
Now I my ODs are off because i cant set the baseline blank for the media. Should I change my method, or should the software/process be adjusted to allow my approach.
Disadvantages of current method.
No ability to use a 0.0 point.
Requires recalibration with changing media formulation.
Shelf life of standards is limited by having to include media.
