I just autoclaved bottles of growth medium with the GL45 caps and tubes in place, and found the tubing floating above the surface, with the ends entirely outside of the liquid… No matter how much I wiggle the cap, swirl the bottle etc, they just float back to the top, where they can’t suck in any liquid.
The density of silicone should be higher than that of water, and the medium isn’t anything special (synthetic minimal medium for yeast with 2% glucose) so it shouldn’t be particularly dense.
Has anyone experienced this?
Any thoughts as to how I can best keep the ends of the tubing all the way at the bottom of the bottles?
