Temperature reading accuracy

Hi everyone,

We used a standard glass thermometer to measure the temperature of the Pioreactor 20 mL v1.1 models running with thermostat automation set to 37C. The thermometer measurements showed 35.5C for P02 & P06, and 35C for P05. However, as you can see in the plot attached below, the reported values were 2-3C lower than the measured values (~32c for P02 & P06, and ~34C for P05).

In addition, when the themometer was used, the temperature in two reactors dropped quite a lot. The configuration setup has not been adjusted. Do you think changing the K values can help to solve the issue?

Did anyone else measured the temperature levels of their media or experienced similar issue?

Is this using the 30ml of culture in 60ml vials? (i.e. the same set up as in Thermostat target cannot be reached ?)

Yes, though I have also encountered the same issue while running outside the laminar flow bench with 15 mL cultures in 20 mL vials with Pioreactor 20 mL v1.1 setup, so the case is not strictly limited with Thermostat target cannot be reached.

With 30ml liquid in 60ml vials, our temp. algorithm will likely be off - though it’s hard to say how much. The algorithm uses a model based on data from standard 20ml vials in a Pioreactor 1.1, with ~10ml of liquid. Even with these conditions, some variance is expected: the heater comes on and off, and you’ll see periods of temperature changes.

That being said, it’s probably time to refresh the algorithm!
