Upgrade 40 mL pioreactors

Would you recommend using different OD calibration curves when changing from 20 mL pioreactors to 40 mL ones?

Mmm, in theory you should (since the position of the LED and PD has changed slightly), but I bet you could get away with not re-calibrating.
